Scientists identify regenerative checkpoint that limits muscle repair

Good news!

"Researchers ... have identified a molecular mechanism that constrains skeletal muscle regeneration and myofiber repair, a finding that could lead to improved treatment for conditions like muscular dystrophy and severe injury.

To repair muscle, muscle cells, or myocytes, fuse to one another. But the molecular pathways that signal this cell-to-cell fusion have remained poorly defined. ...

The team ... found that platelet-derived growth factor receptor beta (PDGFRb), a receptor protein located in cell membranes, is a key modulator of myocyte function in adult muscle cells. ...

Through in vitro and in vivo experiments, they found that genetic deletion of PDGFRb enhanced muscle regeneration and increased myofiber size, whereas PDGFRb activation impaired muscle repair. ..."

From the abstract:
"Muscle cell fusion is critical for the formation and maintenance of multinucleated myotubes during skeletal muscle development and regeneration. However, the molecular mechanisms directing cell-cell fusion are not fully understood.
Here, we identified platelet-derived growth factor receptor β (PDGFRβ) signaling as a key modulator of myocyte function in adult muscle cells.
Our findings demonstrated that genetic deletion of Pdgfrb enhanced muscle regeneration and increased myofiber size, whereas Pdgfrb activation impaired muscle repair.
Inhibition of PDGFRβ activity promoted myonuclear accretion in both mouse and human myotubes, whereas PDGFRβ activation stalled myotube development by preventing cell spreading to limit fusion potential.
Furthermore, PDGFRβ activity cooperated with TGF-β signaling to regulate myocyte size and fusion. Mechanistically, PDGFRβ signaling required STAT1 activation, and blocking STAT1 phosphorylation enhanced myofiber repair and size during regeneration. Collectively, PDGFRβ signaling acts as a regenerative checkpoint and represents a potential clinical target to improve skeletal muscle repair."

New York governor clears path for robotaxis everywhere except for New York City. Really!

One can only hope, the legislators of NY are smarter than Governor Hochul (D, age 67)!

Is this some kind of a horse trading deal by the Dimocratic Party Governor?

"New York Governor Kathy Hochul plans to introduce legislation that would effectively legalize robotaxis in the state — except for its most populous metropolis: New York City. 

Hochul, who made the comments Tuesday during her State of the State address, said the legislation would advance the next phase of the state’s autonomous vehicle pilot program.

Details on the proposed legislation and when it might be released are thin. However, there are some hints contained within a document that outlines an array of proposals and promises Hochul made in her State of the State address. 

Among them is language to expand the state’s existing AV pilot program to allow for “the limited deployment of commercial for-hire autonomous passenger vehicles outside New York City.”  ..."

Two Sanctuary States, New York and Illinois have Released Nearly 9,000 Criminal Aliens with ICE Detainers Since January

Incredible! Insane! Will somebody be held accountable? Don't hold your breath!

Just two of those states, New York and Illinois, have released nearly 9,000 criminal aliens from jails and prisons since January 20, defying Immigration and Customs Enforcement (ICE) detainers to deport them. Among the offenders protected by the sanctuary policies are murderers, terrorists, sexual predators and kidnappers, the Department of Homeland Security (DHS) confirms this month.
Almost 7,000 criminal aliens flagged by ICE were released from custody in the state of New York during the roughly 10-month period examined by DHS. Illinois released more than 1,700 criminal aliens from jails and prisons throughout the state rather than turn them over to federal authorities for removal. The failure to honor ICE detainers has led to “numerous illegal alien murderers, rapists, and pedophiles being released” back into sanctuary communities, Homeland Security officials have established. ..."

NYC Mayor Eric Adams Declares Himself a Maccabee at Chanukah Farewell

US Supreme Court said “No” to the State of New York’s attempt to force an Amish community to vaccinate their school-age children.

Good news! Bravo!

The Amish and Mennonite communities of America are one of the reasons for American Exceptionalism!

"... For religious reasons, the Amish educate their children in private Amish schools and have done so for hundreds of years without vaccinations.

And for over 50 years, New York accommodated their religious belief by granting religious exemptions from the State’s school vaccination requirements. But in 2019, the State repealed its religious exemption for vaccine mandates for school children.

In 2022, the State imposed catastrophic penalties on the Amish community in its attempt to force them to comply.

New York's Medical Aid in Dying Bill a Shameful Act of False Compassion. Really!

Besides abortion (beginning of human life), assisted suicide (end of human life) are two among the biggest or most controversial hot button issues at least for the past 50 years or so in Western societies.

Here are some brief comments/questions:
  1. The right to individual suicide is undeniable when an individual voluntarily and without external influence/pressure chooses/consents to do so. This does not preclude or prevent any attempts to save the individual before, during or after suicide attempt. It does not prevent any loving care of the individual by family or friends.
  2. Is there any requirement to assist such an individual with their suicide attempt?
  3. Assisted suicide may prevent unnecessary suffering while committing or during suicide. Assisted suicide allows for consultation and a more peaceful passing away then perhaps otherwise. 
  4. Is it legitimate or morally acceptable if an older, perhaps not very healthy,  individual chooses suicide to avoid becoming a burden to others? It is in accordance with the individual right to suicide and dignity of that individual.
  5. Should it be limited only to mentally fit adults? What about seriously suffering children?
  6. Should it be limited only to terminally ill or end of life individuals?
  7. Should doctors be actively involved in assisted suicide (e.g. administering injections etc.)? Or should doctors only provide means (e.g. pills) to commit suicide to the individual who then chooses to apply the means.
  8. Active, direct involvement in assisted suicide by doctors is in violation of their Hippocratic Oath (do no harm). Doctors are supposed to prolong and improve healthy lives not end lives.
  9. I believe it is empirically demonstrated that originally such laws have been passed to assist suicide limited only to terminally ill or end of life individuals, but later these laws were amended to include other individuals or all individuals.
Here is what the government of the Netherlands says about euthanasia of children:
"Euthanasia and minors
A child may request euthanasia from the age of 12. However, the following additional requirements apply:
the child must be capable of assessing and understanding what is best for them in their situation;
if a child is aged 12-15, euthanasia can only be carried out with the consent of the child’s parent or parents, or the child’s guardian;
if a child is aged 16 or 17, the parent(s) or guardian must be consulted in the decision-making process, but their consent is not required. ..."
Is the Netherlands going to far with euthanasia? Do the people of the Netherlands largely agree with how euthanasia is legalized?

"... On Monday, the New York state Senate passed legislation to legalize assisted suicide by a vote of 35 to 27. With just six Democrats joining Republicans in opposition, the Medical Aid in Dying Act now moves one step closer to becoming law and making New York the 12th state in the nation to legalize assisted suicide. ...

This bill has some of the loosest safeguards in the country, stripping away even the bare minimum protections seen in other states.

There is no mandatory waiting period. No requirement for in-person evaluations. Mental health screening is only required if a doctor suspects impaired judgment, which, given that evaluations can be done virtually, opens the door wide to inaccurate diagnoses. New York’s bill allows for assessments over Zoom and no follow-up. There is no requirement for physicians or pharmacists to report cases to the state.

Supporters insist the law is only for those near death, those with terminal diagnoses who are suffering. But we’ve heard this all before. We’ve seen what happens when assisted suicide becomes legal. In countries such as Canada and the Netherlands, eligibility quickly expanded beyond the terminally ill.

In the Netherlands, assisted suicide accounts for nearly 6% of all deaths, and proposals have been floated to allow otherwise healthy seniors over the age of 75 to request assisted suicide after six months of counseling.

In Canada, officials have documented cases of patients seeking death not due to pain, but because of poverty, loneliness, or fear of becoming a burden to others. ...

But there is another cost to this bill. The erosion of something deeply precious; namely, our culture of care. ...

They bathed their loved ones, managed pain medication, and sat with them through the long, quiet hours of dying.

Many were holding down jobs or raising kids at the same time. But they still chose to do it. To serve. To love. ..."

A common labor inducing drug, applied topical to neck, restores youthful function in aging mice brains by removing waste from the brain

"... In 2012 Danish neuroscientist, Maiken Nedergaard first described the system that uses cerebrospinal fluid (CSF) to remove waste from the brain and termed it the glymphatic system.
Now, Nedergaard and her colleagues have looked deeper into the glymphatic system, focusing on lymph vessels called lymphangions. These are a series of tiny pumps in the neck that are responsible for moving dirty CSF out of the brain and into the lymph system where it ultimately reaches the kidneys to be processed. ...
Using advanced particle tracking in mice models, they found that as the rodents aged, the contractions in these pumps decreased. As a result, they found that older mice had 63% less dirty CSF flowing out of their brains compared to younger mice ...
Nedergaard focused on the fact that lymphangions are lined with smooth muscle cells. So they looked to a drug called prostaglandin F2α, which works on these types of cells and is commonly used to induce labor in pregnant women. ... administering the drug to the elderly mice with a topical cream got the pumps working again to the point that the team saw the flow of dirty CSF out of the brain return to the same level of efficiency seen in younger mice. ..."

"Alzheimer’s, Parkinson’s, and other neurological disorders can be seen as “dirty brain” diseases, where the brain struggles to clear out harmful waste. Aging is a key risk factor because, as we grow older, our brain's ability to remove toxic buildup slows down. However, new research in mice demonstrates that it’s possible to reverse age-related effects and restore the brain’s waste-clearing process. ..."

"Cervical lymphatic vessels (cLVs) have been shown to drain solutes and cerebrospinal fluid (CSF) from the brain. However, their hydrodynamical properties have never been evaluated in vivo. Here, we developed two-photon optical imaging with particle tracking in vivo of CSF tracers (2P-OPTIC) in superficial and deep cLVs of mice, characterizing their flow and showing that the major driver is intrinsic pumping by contraction of the lymphatic vessel wall. Moreover, contraction frequency and flow velocity were reduced in aged mice, which coincided with a reduction in smooth muscle actin expression. Slowed flow in aged mice was rescued using topical application of prostaglandin F2α, a prostanoid that increases smooth muscle contractility, which restored lymphatic function in aged mice and enhanced central nervous system clearance. We show that cLVs are important regulators of CSF drainage and that restoring their function is an effective therapy for improving clearance in aging."
