A Tweak to Immune Cells Reverses Aging in Mice

Amazing stuff! Good news!

"... But the mechanisms behind age-related inflammation are not well understood. In a study published today (January 20) in Nature, researchers show that older immune cells have a defect in metabolism that when corrected in a mouse model of Alzheimer’s disease can decrease inflammation and restore cognitive function. ...
COX-2 activation is the first step in the production of a lipid called prostaglandin E2 (PGE2), which can bind to one of its receptors, EP2, on immune cells and promote inflammation. To plug up the pathway, [researchers have] shown that deleting the EP2 receptor in mouse macrophages and brain-specific microglia—the cells normally responsible for detecting and destroying immune invaders and cellular debris—reduces inflammation and increases neuronal survival in response to both a bacterial toxin and a neurotoxin. ... "

"... However, the underlying mechanisms that initiate and sustain maladaptive inflammation with ageing are not well defined. Here we show that in ageing mice myeloid cell bioenergetics are suppressed in response to increased signalling by the lipid messenger prostaglandin E2 (PGE2), a major modulator of inflammation. In ageing macrophages and microglia, PGE2 signalling through its EP2 receptor promotes the sequestration of glucose into glycogen, reducing glucose flux and mitochondrial respiration. This energy-deficient state, which drives maladaptive pro-inflammatory responses, is further augmented by a dependence of aged myeloid cells on glucose as a principal fuel source. In aged mice, inhibition of myeloid EP2 signalling rejuvenates cellular bioenergetics, systemic and brain inflammatory states, hippocampal synaptic plasticity and spatial memory. ...
Our study suggests that cognitive ageing ... can be reversed by reprogramming myeloid glucose metabolism to restore youthful immune functions."

Catching cancer in the act of metastasis

Good news! This could be a breakthrough!  Cancer is history!

"... The exact moment of metastasis is fleeting, lost in the millions of divisions that take place in a tumor. “These events are typically impossible to monitor in real time,” ...
Now, researchers led by Weissman, who is also an investigator with the Howard Hughes Medical Institute, have turned a CRISPR tool into a way to do just that. ..."

"Detailed phylogenies of tumor populations can recount the history and chronology of critical events during cancer progression, such as metastatic dissemination. We applied a Cas9-based, single-cell lineage tracer to study the rates, routes, and drivers of metastasis in a lung cancer xenograft mouse model. We report deeply resolved phylogenies for tens of thousands of cancer cells traced over months of growth and dissemination. ...

Harvard researchers sequence Sapria genome

Amazing stuff!

"When it comes to parasites, these plants are the stuff of nightmares.

Called Rafflesiaceae, they have no roots, stems, or leaves of their own. For most of their lifespan they are invisible, living only as a small necklace of cells inside the woody vines of their host. Then, without warning — like the creature in the movie “Alien” — they burst out to bloom some of the largest flowers in the world. Their pungent smell of rotting meat or fruit attracts the carrion flies that help pollinate these plants, allowing them to seed and spread to another unsuspecting host, restarting the whole cycle. Rafflesiaceae represent the most extreme form of parasitism, known as endoparasitism, in which the organism is completely dependent on its host for all nutrients. ...
On Jan. 22 in Current Biology, a team of Harvard-led researchers presented the most complete genome yet assembled of one of the major Rafflesiaceae lineages, Sapria himalayana. ...
The genetic analysis revealed an astonishing degree of gene loss and surprising amounts of gene theft from its ancient and modern hosts. ..."

Stacker lets anyone transform spreadsheets and databases into web apps

Sounds like an interesting and useful business proposition!

"Low-code and no-code development tools were one of the big trends to emerge last year, with Google snapping up enterprise-focused AppSheet and Amazon’s AWS launching Honeycode. This trend seems to be accelerating into 2021, with no-code development platform Webflow raising $140 million at a $2.1 billion valuation earlier this month and newcomers such as automated web testing platform Reflect bolstering their coffers. ...
The fact that almost every company is now a software company on some level means this trend will continue to grow.
According to some reports, the global low-code development market is expected to generate $187 billion in revenue annually by 2030, up from $10.3 billion in 2019 ...
Stacker serves as an “app layer” on top of the spreadsheet, providing a friendly interface for the raw data alongside added features such as user logins, access controls, and more. ...
to set his platform apart by letting people without any coding knowledge create web apps for both internal and customer-facing scenarios. When American Movies were still great!

Today, an unusual marketing campaign from the Epoch Times landed in my email inbox. I will repeat it here since I am also a great fan of the Forrest Gump movie of 1994:

"Recently, a friend told me that she’s never seen Forrest Gump before—which is a crime in my house. So I made her watch the movie with me (spoilers ahead).

In light of recent events, this movie has taken on a new meaning for me.

Though Forrest and Jenny started “like peas and carrots,” each of them soon got swept up by a different one of America’s 20th-century cultural currents.

Forrest’s path was classically American: going to college, playing football, serving his country, starting his own business, and going to church.

Jenny, on the other hand, dabbles in counterculture: dropping out of college, becoming a beatnik and then a hippie, joining the anti-war protests, indulging in the sex, drugs, and rock n’ roll of the ‘70s.

In these two characters, I saw a reflection of today’s America—with one side supporting the American tradition and the other opposing it.

Jenny’s path finds her used and abused multiple times, first as a sex worker and then as a girlfriend. While she experiences momentary freedom as a hippie, she eventually devolves into a groupie who steals from the musicians she sleeps with and hits her lowest point when she attempts suicide.

But Forrest never stops loving Jenny, no matter how much their paths deviated from each other. He gets rejected by her multiple times but remains there for her through everything—never imposing, never selfish, never angry, always altruistic.

It’s precisely because of Forrest’s love that Jenny is able to find happiness in the end, and it’s a kind of love that I think we can use more of in our society today.

2020 has shown me the destructive power of the forces that are working against our traditional values. Coming from a communist country, I know that these forces don’t just destroy their enemies: they also destroy the same people who advocate for them.

Many people around me don’t truly understand these forces, however, and they’re people I care for deeply. Like Jenny, they were attracted to the prospect of freedom that these forces offered, and like Jenny, they’re unable to see the destruction that these forces could cause to a person.

It's not easy, but I think that the best thing I can do for them is to be their Forrest. Even though they may seem to be confident in their decisions now, I have to be there for them with compassion and patience. I have to love them simply and selflessly; I have to stay firm in my own convictions and not give up.

For history tells me that one day, they might regret their decisions. When that day comes, they’re going to need someone there to help bring them back.

The end of liberal diplomacy?

Mostly very recommendable!

"[President] Trump inadvertently made an important point: the emperor—the prevailing diplomatic approach—has no clothes. ... Those norms are inextricably linked to the liberal world order that emerged after World War II. The ‘responsibility to protect’ (R2P) doctrine—the world’s commitment, adopted unanimously by the UN General Assembly in 2005, to protect populations from genocide, war crimes, ethnic cleansing and crimes against humanity—exemplifies this liberal diplomacy. ... But, in the past couple of decades, it has been all downhill for this vision. In Libya—the first case where the UN Security Council authorised a military intervention based on R2P—UN envoys come and go, but the country’s future is decided by foreign powers acting unilaterally. And, with the Security Council deadlocked, R2P hasn’t been invoked to justify military intervention since, despite several notable mass atrocities perpetrated by people’s own governments.

The repeated failure of the UN’s collective security system can be partly attributed to the decline of the liberal world order itself. Long before Trump, America had become increasingly reluctant to act as the order’s guarantor (in Libya, President Barack Obama vowed that the US would ‘lead from behind’). Add to this Russia’s aggressive revisionism, China’s abandonment of its ‘peaceful rise’ and the European Union’s preoccupation with its own survival. ...
But many of the world’s biggest diplomatic challenges—from the Israeli–Palestinian conflict to the dispute over Western Sahara—predate these factors. Even at its peak, liberal diplomacy could not resolve them, not least because it too often treated statecraft as an expressive art, detached from an ever-changing reality.

Consider the fight over Western Sahara—Africa’s longest-running territorial dispute. In 1975, with Spain poised to cede control of the territory, the International Court of Justice rejected Morocco’s claim to it, and ruled that the local people, the Sahrawis, were entitled to self-determination. But Morocco quickly invaded and annexed the territory. ...
For example, the recent eruption of the decades-old conflict between Armenia and Azerbaijan over the enclave of Nagorno-Karabakh ended with a Russia-brokered deal that legitimised Azerbaijan’s annexation of a significant amount of territory. Russian peacekeeping forces were dispatched to enforce the deal. The UN was nowhere to be found. ..."

WHO Changes CCP Virus Test Criteria in Attempt to Reduce False Positives

This is hot stuff! E.g. for some time now there were doubts published on the over expression issue of the PCR (Polymerase chain reaction) test! Myself, I have had some doubts about the high number of new cases and, in particular, death cases beginning in July of 2020. What took the WHO so long to correct the issue!

"The World Health Organization (WHO) has cautioned experts not to rely solely on the results of a PCR test to detect the [SARS-CoV-2] virus.
In updated guidance published on Jan. 20, the WHO said that lab experts and health care practitioners should also consider the patient’s history and epidemiological risk factors alongside the PCR test in diagnosing the ... virus.
The new guidance could result in significantly fewer daily cases.
Most PCR assays are indicated as an aid for diagnosis, therefore, health care providers must consider any result in combination with timing of sampling, specimen type, assay specifics, clinical observations, patient history, confirmed status of any contacts, and epidemiological information,” the guidance says. ...

Scientists and physicians have raised concerns for many months of an over-reliance on and a misuse of the PCR test as a diagnostic tool since it can’t differentiate between a live infectious virus from an inactivated virus fragment that is not infectious.

Additionally, the high cycle threshold values of most PCR tests—at 40 cycles or higher—increases the risk of false positives. ...
And no matter how many times an [same] individual is tested, each positive test is counted as a separate case. ..."

Jane Dieulafoy, a 19th-century French archaeologist, broke the law by wearing pants.

Let me guess, this is one of those many articles that try to establish the contributions of women to science, literature etc. Like many other people, I do not remember that I have ever heard or read about Jane Dieulafoy.

Like famous men had often very supportive wives or female companions without them their lives and accomplishments would have been very different, the same can be assumed for famous women.

What seems to be clear though is that her longtime husband fully supported his wife's endeavors and her expression of masculinity/cross-dressing/tomboy. Her archaeological exploits were accomplished with her husband together. One may speculate that without her very supporting husband, her life would have been very different.

This lady certainly had an unusual life (excerpts are from Wikipedia) plus my comments (in blue):
  1. "That same year [1870], the Franco-Prussian War began. Marcel [her husband] volunteered, and was sent to the front. Jane accompanied him, wearing a soldier's uniform and fighting by his side."
  2. "With the end of the war, Marcel was employed by the Midi railways, but during the next ten years the Dieulafoys would travel in Egypt and Morocco for archaeological and exploration work. Jane did not keep a record of these journeys. Marcel became increasingly interested in the relationship between Oriental and Western architecture, and in 1879 decided to devote himself to archaeology."
  3. "During her travels abroad, Jane Dieulafoy preferred to dress in men's clothing and wear her hair short, because it was otherwise difficult for a woman to travel freely in a Muslim country. She had also dressed as a man when she fought alongside Marcel Dieulafoy during the Franco-Prussian war and she kept dressing in men's clothing when she came back in France. This was against the law in France at the time, but when she returned from the Middle East she received special "permission de travestissement" from the prefect of police."
One may also safely speculate that this prefect of police was a male at that time who issued this permission to cross-dress to her! One may further speculate that women in male clothing at the time were perhaps not so rare as generally believed.
  4. "It is difficult to determine Dieulafoy's motives for choosing to wear men's clothing. She wrote, "I only do this to save time. I buy ready-made suits and I can use the time saved this way to do more work" but, given that she includes many characters who cross-dress in her fiction ... she was clearly fascinated with the subject of cross-dressing and her motives appear to be more complex than mere convenience."
  5. "Dieulafoy considered herself an equal to her husband, but was also fiercely loyal to him. She was opposed to the idea of divorce, believing it degraded women."
Well that may disappoint some feminists!
  6. "During the First World War, she petitioned for allowing women a greater role in the military."
Gender equality certainly means that women like men should be e.g. drafted into military service and into combat roles. Many feminists most certainly do not like this idea to bring women in harms way, they rarely demand military service equality to the best of my knowledge!

The President’s Advisory 1776 Commission Report

Unfortunately, I have had no time yet to read it! However, I bet most of its content is to my liking!

None other than Larry P. Arnn (president of Hillsdale College) was the chair of this commission. Victor Davis Hansen was another member of it!

"... The Commission issued its 1776 Report this past Monday. The report calls for a return to the unifying principles stated in the Declaration of Independence. It quotes the greatest Americans, black and white, men and women, in devotion to these principles. It acknowledges the many ways we have fallen short of them even as it celebrates, following Abraham Lincoln, the influence for good that they exercised to the benefit of all. It acknowledges the way we fall short of them today and argues that it is only by returning to them that our current evils can be corrected. It calls for a civics education that fosters reverence for these principles, beginning with an accurate and honest teaching of American history. It is not a partisan document. ..."

A few quickly picked excerpts:
  1. "The Declaration of Independence is the ring-bolt to the chain of your nation’s destiny; so, indeed, I regard it. The principles contained in that instrument are saving principles. Stand by those principles, be true to them on all occasions, in all places, against all foes, and at whatever cost."
    Frederick Douglass"
  2. "History underscores the overwhelming importance of religious faith in American life, but some today see religious practice and political liberty to be in conflict and hold that religion is divisive and should be kept out of the public square. The founders of America held a very different view. They not only believed that all people have a right to religious liberty but also that religious faith is indispensable to the success of republican government. “The God who gave us life, gave us liberty at the same time,” Thomas Jefferson once wrote. “The hand of force may destroy, but cannot disjoin them.” The idea that faith sustains the principles of equality and natural rights is deeply rooted in American society and proven through human experience."
  3. “Of all the dispositions and habits which lead to political prosperity, Religion and morality are indispensable supports,” George Washington observed in his Farewell Address. “In vain would that man claim the tribute of Patriotism, who should labor to subvert these great Pillars of human happiness, these firmest props of the duties of Men and citizens.” He went on to warn: "Let us with caution indulge the supposition, that morality can be maintained without religion. Whatever may be conceded to the influence of refined education on minds of peculiar structure, reason and experience both forbid us to expect that National morality can prevail in exclusion of religious principle."
  4. [Racial, sexual, and gender group] Identity politics divide Americans by placing them perpetually in conflict with each other. This extreme ideology assaults and undermines the American principle of equality in several key ways.

"Living things leave behind tell-tale signs of their existence: fossilized bones, DNA, the chemical byproducts of metabolism. Living things on Earth, that is. There currently exists no predictive theory to completely guide astrobiologists searching for life beyond our planet, where the chemical signatures of life, and the geo- and atmospheric chemistries under which it evolved, might look quite different from Earth. In early November [2020], NASA’s Astrobiology Program launched the new Interdisciplinary Consortia for Astrobiology Research (ICAR), supporting eight teams focusing on specific outstanding questions in astrobiology. ..."

The hole story of how cage molecules could transform synthesis and separation

Amazing stuff! This approach might have great potential! This is a very recommendable overview article!

"... Molecules twist and fold into unusual shapes to fit inside a nanosized container, concentration increases by factors of up to 10,000, and unstable intermediates become less unstable as they are shielded from the outside world. At the molecular scale, interactions between container and contained molecules become ‘prearranged, prolonged and private’ ...
After 35 years of research, chemists are now starting to unlock the potential of confinement by designing molecular cages that fold up long molecules into huge macrocycles, or creating cavities that can separate deuterium from hydrogen. In the future, confined spaces could rival enzymes in their ability to catalyse reactions or protect unstable species, turning them into useful reagents. ...
One of the most complex transformations ever carried out inside a cage was reported by Ken Raymond, Robert Bergman and Dean Toste, a long-time supramolecular chemistry collaboration at the University of California, Berkeley, US. They used a gallium coordination cage, nicknamed Raymond tetrahedron, to catalyse a three-component coupling reaction.6 More than a simple catalyst, albeit one that increases yields up to 12-fold, the tetrahedron also inverts the reaction’s stereoselectivity. In bulk solution, the product created is the opposite stereoisomer to the one generated inside the cage."

The Inaugural Address of President Coolidge That Warned of the Dangers of ‘Legalized Larceny’ by Lawrence Reed

Recommendable! President Coolidge is probably one of the most underrated presidents of the last 120 years or so. The 46th President will probably be no match for President Coolidge or he will even go down as one of the worst socialists since FDR!

"... History teaches endless lessons whether people want to learn them or not. Its pages instruct us painfully that the two greatest dangers from government are mission creep and creeps on a mission. The last thing you would ever hear from the lips of Calvin Coolidge were arrogant pretensions to knowledge or grand plans to “fundamentally transform” America. ..."

Genetic differences between monozygotic twins

Amazing stuff coming again out of tiny Iceland!

"Despite the important role that monozygotic twins have played in genetics research, little is known about their genomic differences. Here we show that monozygotic twins differ on average by 5.2 early developmental mutations and that approximately 15% of monozygotic twins have a substantial number of these early developmental mutations specific to one of them. ..."

"... Rather than having exactly the same DNA sequences, twins start accumulating genetic variation from the earliest stages of development, researchers at Iceland-based company deCODE genetics [founded in 1996] found, meaning that one twin harbors variants that aren’t present in the other. ...
The new study focuses specifically on mutations that occur as or before embryos form from the mass of cells inside the blastocyst, a structure that implants in the uterine wall. During this stage of development, this inner cell mass can split to form two separately developing embryos.
Drawing on deCODE’s genetic databases to analyze variation in multiple cells sampled from 381 monozygotic twin pairs and their immediate family members, the researchers pinpointed a number of mutations that arose during this stage of development in just one member of each pair, meaning that at birth, so-called identical twins may already differ genetically from each other.

China’s surging private space industry is out to challenge the US

Recommendable! The acceleration of the space race appears imminent! I believe India is not very far behind and so on.

"China’s space program might have been slowed by the pandemic in 2020, but it certainly didn’t stop. The year’s [2020] highlights included sending a rover to Mars, bringing moon rocks back to Earth, and testing out the next-generation crewed vehicle that should take taikonauts into orbit ...
But there were a few achievements the rest of the world might not have noticed. One was the November 7 launch of Ceres-1, a new type of rocket that, at just 62 feet in height, is capable of taking 770 pounds of payload into low Earth orbit. The launch sent the Tianqi 11 communications satellite into space. ... Ceres-1, however, wasn’t built and launched by China’s national program. It was a commercial rocket—only the second from a Chinese company ever to go into space. And the launch happened less than three years after the company was founded. ...
China is set to build a new space station later this year and will likely attempt to send its taikonauts to the moon before the decade ends. ...
the [Chinese] government thinks its commercial space sector can be complementary to the state ...
The space industry is undergoing a renaissance thanks to two big trends spurred by the commercial industry: we can make satellites for less money by making them smaller and using off-the-shelf hardware; and we can also make rockets for less money, by using less costly materials or reusing boosters after they’ve already flown ...
In 2014, a year after Xi Jinping took over as the new leader of China, the Chinese government decided to treat civil space development as a key area of innovation ...
As a result, there are now 78 commercial space companies operating in China, according to a 2019 report by the Institute for Defense Analyses. More than half have been founded since 2014, and the vast majority focus on satellite manufacturing and launch services. ..."

Did Public Schooling Fail Us?

Recommendable! Food for thought!

Government shall not run public schools and the education of our children! Largely privatize public schooling! Public schooling of most children was not established until the late 19th century. Before that children were already reasonably educated without Big Government! We the people are very well capable of organizing the education of our children!

"... We are told that public schooling is the “cornerstone” or “bedrock” of American democracy, and anything departing from it would cause unity and rule by the people to crumble. ...
The notion that “common” schooling is crucial to democracy has some illustrious early propagators. Thomas Jefferson – generally not a proponent of energetic government – was perhaps the most famous Founder to champion public schooling, calling for a basic, publicly funded schooling system in Virginia. He wrote in 1786, “I think by far the most important bill in our whole code is that for the diffusion of knowledge among the people. No other sure foundation can be devised for the preservation of freedom, and happiness.” He was not alone in this belief, with John Adams and others also asserting the importance of widespread education delivered, at least partially, by public schools.
Despite the enthusiasm for public schooling among some founding luminaries, little was established in their lifetimes. ...
It took several decades after [Horace] Mann’s [1848] crusade to solidify public schooling as the educational norm, but it eventually came to dominate. The earliest, readily available data we have on where children have gone to school dates back to 1889, and they consistently show roughly 90 percent of students attending public schools. We also see more children spending increasing amounts of time in school as time passes: from about 65 percent of 5 to 17‐year‐olds spending roughly 78 days in school in 1869, to more than 94 percent spending 167 days today. ...
The Capitol riot seems like prima facie evidence of a dissolution of social cohesion and democratic values, especially coming off years of highly contentious elections, demonstrations against police brutality that sometimes spun off destruction, “cancel culture,” and more. Deeper analyses back up this seemingly obvious state of affairs. These include non‐negligible support for unilateral presidential rule or even a military coup under certain conditions; findings of markedly increased demonization of members of opposing political parties since the late 1960s; partisan affiliations being seen as legitimate negative factors to use in hiring decisions; declining trust in numerous American institutions, including government; and declining generalized trust, with the share of General Social Survey respondents agreeing that “most people can be trusted” dropping from 46 percent in 1972 to 21 percent in 2018. ...
Clearly, widespread public schooling has not prevented the decay of social cohesion and democratic norms. Probably because it never had the power to do so. The education system cannot force people to like one another or democracy, nor can it maintain those things if people turn against them. Democratically controlled schooling is more likely to reflect decrepit attitudes than change them. ..."
