In the new study, the researchers used cryo-electron microscopy—a cutting-edge technique that reveals the locations of atoms within a frozen protein sample—to analyze the precise arrangement of molecules that form the Flycatcher1 protein channel in Venus flytrap plants. They found that Flycatcher1 is, in many ways, similar to bacterial MscS proteins—seven groups of identical helices surrounding a central channel. But, unlike other MscS channels, Flycatcher1 has an unusual linker region extending outward from each group of helices. Like a switch, each linker can be flipped up or down. When the team determined the structure of Flycatcher1, they found six linkers in the down position, and just one flipped up. ..."

"Flycatcher1 (FLYC1), a MscS homolog, has recently been identified as a candidate mechanosensitive (MS) ion channel involved in Venus flytrap prey recognition. FLYC1 is a larger protein and its sequence diverges from previously studied MscS homologs, suggesting it has unique structural features that contribute to its function. Here, we characterize FLYC1 by cryo-electron microscopy, molecular dynamics simulations, and electrophysiology. Akin to bacterial MscS and plant MSL1 channels, we find that FLYC1 central core includes side portals in the cytoplasmic cage that regulate ion preference and conduction, by identifying critical residues that modulate channel conductance. Topologically unique cytoplasmic flanking regions can adopt ‘up’ or ‘down’ conformations, making the channel asymmetric. ..."

We demonstrated that recruitment of Sca-1+ monocytes into lymphoid organs during Salmonella Typhimurium (STm) infection disrupted pre-existing germinal center (GC) reactions. GC responses induced by influenza, plasmodium, or commensals deteriorated following STm infection. GC disruption was independent of the direct bacterial interactions with B cells and instead was induced through recruitment of CCR2-dependent Sca-1+ monocytes into the lymphoid organs. "Photothermal therapy – a cancer treatment based on laser irradiation of injected gold nanorods – provides high tumour targeting accuracy with low toxicity. Now researchers in China have developed a new nanoplatform that completely eliminates solid liver cancer tumours in mice following a single five-minute light dose and without toxic side effects.

The treatment works by injecting gold nanorods directly into a tumour and then irritating them with near-infrared (NIR) laser light. This causes the nanorods to heat up and kill surrounding cancer cells. Photothermal therapy alone, however, can fail due to incomplete tumour ablation, particularly for larger lesions. This is mainly due to the limited penetration depth of light into tissue and the nanorods not fully permeating throughout the tumour.

To address these issues, the researchers deposited clusters of palladium (Pd) on the ends of gold (Au) nanorods to create heterogeneous Pd-Au nanorods. To improve biocompatibility, they modified the nanorod surfaces with bovine serum albumin (BSA). The introduction of Pd nanoclusters improves the photothermal therapy in three unique ways[:]

[1)] the BSA-Pd-Au nanorods absorb light at longer wavelengths than standard gold nanorods, extending the maximum absorption wavelength from 950 nm to 1050 nm. In this NIR-II region, light penetrates deeper into tissue and can be used at higher maximum skin exposures.

[2)], the nanorods can be used to activate Pro-5-Fu, a prodrug of the chemotherapy drug 5-fluorouracil (5-Fu). By injecting Pro-5-Fu intraperitoneally, then synthesizing 5-Fu in situ at the site of the BSA-Pd-Au nanorods, the potential for damaging healthy cells is minimized.
[3)], the nanorods convert hydrogen peroxide into harmful reactive oxygen species. This so-called chemodynamic therapy (CDT) helps eliminate any residual tumour cells. ..."

"Researchers at the Weizmann Institute of Science, in collaboration with the National Cancer Institute, USA, and other institutions, have now revealed a mechanism that accounts for the spread of a particularly aggressive type of breast cancer.

The study began with a large-scale computational analysis of data from several databases, including information on patient survival and the genes that are overly active in fast-spreading cancers. The scientists came up with a list of some 20 genes, out of about 25,000, that the most aggressive breast cancers appeared to need in order to spread. The top two are genes that play a role in cell division, and they already serve as targets for anticancer drugs. But the third puzzled researchers: It encoded a protein called NUP93, one of the numerous components of the pores, or tunnels of sorts, in the envelope enclosing a cell’s nucleus. ...
The researchers found that NUP93 was present in abnormally large amounts in breast cancer patients with the poorest survival records. As the disease progressed, the genomes of some of these patients came to contain two or three copies of the gene for NUP93 instead of the usual one. Strikingly, many of these patients had tumors that were not sensitive to estrogen. Such tumors, which make up about a third of all breast cancers, are particularly difficult to treat because they lack the estrogen receptors that are targeted by hormonal anticancer drugs. ...
Next, the scientists unraveled the mechanism by which NUP93 plays its grim role in cancer. Because this protein forms a tunnel in the nuclear membrane, it can allow the passage of shuttle proteins called importins, which, among their cargo loads, deliver growth and migration commands from the cytoplasm into the nucleus and thus to the DNA. In this manner, cancer gets access to the cell’s genome, harnessing it for its own needs, namely, increasing the cell’s ability to migrate and revving it up so as to spread the disease. ..."

A mix of net zero ideologues, naive Nimby’s and renewable energy promoters — often gullibly latching on to Putin’s anti-fracking propaganda — worked tirelessly to stop renewed gas extraction in its tracks. Proponents of the ‘leave it in the ground’ mentality organised protests, stoked public hostility, and made it near-impossible for politicians to reconcile fears with reality. ..."

"... The key to the success of our strategy is to pre-train an expressive GNN at the level of individual nodes as well as entire graphs so that the GNN can learn useful local and global representations simultaneously. We systematically study pre-training on multiple graph classification datasets. We find that naïve strategies, which pre-train GNNs at the level of either entire graphs or individual nodes, give limited improvement and can even lead to negative transfer on many downstream tasks. In contrast, our strategy avoids negative transfer and improves generalization significantly across downstream tasks, leading up to 9.4% absolute improvements in ROC-AUC over non-pre-trained models and achieving state-of-the-art performance for molecular property prediction and protein function prediction."

Working for one of the largest security service companies in the world

Until very recently, I worked part time for one of the top five largest security services companies of the world. It was not Securitas, for which I worked temporarily while studying economics several decades ago. I did not even now that the IT consulting firm Booz Allen Hamilton was also in the securities service business.

As a security officer for this firm, I worked at a very high profile property, a luxury high end shopping mall in Scottsdale, AZ. Perhaps, one of the top five premier shopping malls in Arizona.

The company, I worked for about 2.5 years is particularly lousy in several  respects. Especially towards the end of the Covid-19 pandemic, staffing levels fell to a low of about 50% of normal at this property. The company proved unable to attract new hires, so the situation lasted for over half a year or so. Not only that the last two new hires turned out to have such serious non temporary medical conditions that their work performance was very severely impaired and the other colleagues had to pick up their work.

Despite the extra work imposed on the remaining staff during the Covid-19 pandemic it has not occurred at all to top management to praise the extra work done at the local level and to recognize or reward the loyal and dedicated employees with e.g. bonus checks or what have you. In my 2.5 years with the company, nobody from regional management or other higher management rarely ever showed up on site.

The company does not even offer incentives or rewards like extra bonuses for a job well done! I am not even sure they really track performance at the individual level.

I probably don't have to mention that the hourly pay for part time employees was low like $14 per hour when I started. The company also does not offer to raise the pay even after more than 12 months with the company and a good work record. So it was left to me to finally ask for a pay raise, which was granted, but only to $15 per hour.

In conclusion, the appreciation of this company for its workforce really sucks!

The company also failed to notice that the long time top manager at this site was seriously neglecting his job. He pretended to be some kind of auxiliary manager of the shopping mall. Rarely ever did he care about his employees or mingled with his staff. His office was inside the shopping mall management office. Thus, he rarely visited the break room and supervisor office, which are physically very separated from the shopping mall management office. He foremost and almost exclusively talked only with shopping mall management staff or he chatted with on site city police officers, but not with his security staff. He even went so far to do menial work for the shopping mall management like carrying stuff around. It looked weird! Some of my colleagues wondered what is he actually doing for years all day long other than listening to the radio traffic, but his voice is rarely heard on the radio.

Because of this senior security manager on site, the staff break room, the supervisor office, the common kitchen area, staff restrooms, locker rooms were neglected for months. E.g. broken ceiling lights, broken soap dispenser in the lady's restroom, dirty fridge, poor cleaning and upkeep of the common areas and so on.

There is also a second and third senior manager on this site. While the first manager does barely anything, the second one gets involved in work as needed, but not too often. The third manager on site actually does a lot of work but mostly related to surveillance video recordings to document incidents at the mall and he fills in when needed.

Last, but not least: I don't want to brag, but I was probably one of the top 3 security officers at this high profile site. I worked exclusively interior of the mall only on the busiest days, i.e. Fridays, Saturdays, and Sundays. My service was also very appreciated by the Scottsdale police officers of which two PD officers were always on site on weekends during mall hours. E.g. I had build up over time excellent relationships with many of the mall tenants and their employees. Many of them appreciated my work and for going the extra mile. I had gained their trust so they would sometimes come forward etc. I was usually present and responding at many of the incidences of any kind at the mall.

The End plastic pollution tyranny is coming!

Plastic is turned into something demonic and is compared to a scourge of humanity! The global environmentalist elite wants us citizens to believe that plastic is the next  menace! Much of it is mere propaganda and demagoguery!

For the most part plastic is pretty harmless, but very useful! Plastic can easily be dealt with as long as it is properly disposed of in landfills etc. That some countries lack waste e.g. waste management does not mean all e.g. Western countries need commit to socialist central planning etc. 

We are already developing microbes etc. to dissolve plastics etc. We are also developing plastic that disintegrates/breaks down naturally by itself etc. It is only a matter of time that we have better solutions. There is no need for worldwide massive government intervention! This is a dangerous fantasy by do-gooders and other ideologues!

That too much plastic ends up in the world's oceans is a serious concern, but that should be focused on and addressed as such. Primarily the offenders (some countries more than others) should be targeted!
