Pottery reshapes understanding of Australia's First Nations people

"Aboriginal manufacture and use of pottery was unknown in Australia prior to European settlement, despite well-known ceramic-making traditions in southern Papua New Guinea, eastern Indonesia, and the western Pacific. The absence of ancient pottery manufacture in mainland Australia has long puzzled researchers given other documented deep time Aboriginal exchange networks across the continent and the close proximity of pottery-bearing Lapita and post-Lapita maritime communities in the western Pacific with ocean-going watercraft and sophisticated navigation abilities. We report the oldest securely dated ceramics found in Australia from archaeological excavations on Jiigurru (Lizard Island Group) on the Great Barrier Reef, northeast Australia. Comprehensive radiocarbon dating and Bayesian modelling constrains ceramic deposition to between 2950–2545 cal BP and 1970–1815 cal BP. This timing overlaps with late Lapita and post-Lapita ceramic traditions of southern Papua New Guinea. Geological characterisation of the sherds strongly suggests local manufacture as the vessels belong to three temper and clay groups locally sourced to northeast Australia, and most likely to Jiigurru. The oldest occupation layers date to 6510–5790 cal BP, making Jiigurru the earliest offshore island occupied on the northern Great Barrier Reef. The results demonstrate that northeast Australian First Nations communities had sophisticated canoe voyaging technology and open-sea navigational skills and were intimately engaged in ancient maritime networks, connecting them with peoples, knowledges, and technologies across the Coral Sea region."

"Critical aspects of physiology and cell function exhibit self-sustained ~24-hour variations termed circadian rhythms. In the liver, circadian rhythms play fundamental roles in maintaining organ homeostasis. Here, we established and characterized an in vitro liver experimental system in which primary human hepatocytes display self-sustained oscillations. By generating gene expression profiles of these hepatocytes over time, we demonstrated that their transcriptional state is dynamic across 24 hours and identified a set of cycling genes with functions related to inflammation, drug metabolism, and energy homeostasis. We designed and tested a treatment protocol to minimize atorvastatin- and acetaminophen-induced hepatotoxicity. Last, we documented circadian-dependent induction of pro-inflammatory cytokines when triggered by LPS, IFN-β, or Plasmodium infection in human hepatocytes. Collectively, our findings emphasize that the phase of the circadian cycle has a robust impact on the efficacy and toxicity of drugs, and we provide a test bed to study the timing and magnitude of inflammatory responses over the course of infection in human liver."

"Abstract
Background
Long-term daily use of aspirin reduces incidence and mortality due to colorectal cancer (CRC). This study aimed to analyze the effect of aspirin on the tumor microenvironment, systemic immunity, and on the healthy mucosa surrounding cancer.
Methods
Patients with a diagnosis of CRC operated on from 2015 to 2019 were retrospectively analyzed (METACCRE cohort). Expression of mRNA of immune surveillance-related genes (PD-L1, CD80, CD86, HLA I, and HLA II) in CRC primary cells treated with aspirin were extracted from Gene Expression Omnibus–deposited public database (GSE76583). The experiment was replicated in cell lines. The mucosal immune microenvironment of a subgroup of patients participating in the IMMUNOREACT1 (ClinicalTrials.gov NCT04915326) project was analyzed with immunohistochemistry and flow cytometry.
Results
In the METACCRE Cohort, 12% of 238 patients analyzed were aspirin users. Nodal metastasis was significantly less frequent (p = .008) and tumor-infiltrating lymphocyte infiltration was higher (p = .02) among aspirin users. In the CRC primary cells and selected cell lines, CD80 mRNA expression was increased following aspirin treatment (p = .001). In the healthy mucosa surrounding rectal cancer, the ratio of CD8/CD3 and epithelial cells expressing CD80 was higher in aspirin users (p = .027 and p = .034, respectively).
Conclusions
These data suggested that regular aspirin use may have an active role in enhancing immunosurveillance against CRC."

Atrial fibrillation (AF), the most prevalent heart rhythm disorder, affects millions globally, leading to significant increases in stroke risk, heart failure, and healthcare expenses. These challenges underscore the need for innovative monitoring solutions. Wearable technology, coupled with artificial intelligence, will eventually enable continuous, real-time tracking of heart health and warn users of imminent danger. This paper shows that such a future is not far. Our research introduces a model, WARN, that harnesses R-to-R intervals, the intervals between successive heartbeats, from readily available smartwatches to issue early warnings of AF onset. By leveraging extensive long-term data of individual patients, we expect that WARN can be personalized to significantly improve the prediction horizon, offering a future where many patients might manage AF proactively with as-needed medication rather than routine daily doses, thereby optimizing treatment regimens and improving quality of life.
Summary
Atrial fibrillation (AF), the most prevalent cardiac rhythm disorder, significantly increases hospitalization and health risks. Reverting from AF to sinus rhythm (SR) often requires intensive interventions. This study presents a deep-learning model capable of predicting the transition from SR to AF on average 30.8 min before the onset appears, with an accuracy of 83% and an F1 score of 85% on the test data. This performance was obtained from R-to-R interval signals, which can be accessible from wearable technology. Our model, entitled Warning of Atrial Fibrillation (WARN), consists of a deep convolutional neural network trained and validated on 24-h Holter electrocardiogram data from 280 patients, with 70 additional patients used for testing and further evaluation on 33 patients from two external centers. The low computational cost of WARN makes it ideal for integration into wearable technology, allowing for continuous heart monitoring and early AF detection, which can potentially reduce emergency interventions and improve patient outcomes."

Humans spend a remarkable fraction of their awake time while blinking. Here, we show that eye blinks are not simply a mechanism for refreshing the tear film but act as an information processing stage. By modulating the visual input to the retina, blinks effectively reformat spatial information in the temporal domain, yielding luminance signals that emphasize low-resolution information about the global structure of the visual scene. We show that human observers benefit from these transients and that this perceptual enhancement occurs independently from motor signals associated with blinks. Thus, contrary to common assumption, blinks facilitate—rather than disrupt—visual processing, amply compensating for the loss in stimulus exposure.

... However, blinks also provide luminance transients rich in spatial information to neural pathways highly sensitive to temporal changes. Here, we report that the luminance modulations from blinks enhance visual sensitivity. By coupling high-resolution eye tracking in human observers with modeling of blink transients and spectral analysis of visual input signals, we show that blinking increases the power of retinal stimulation and that this effect significantly enhances visibility despite the time lost in exposure to the external scene. We further show that, as predicted from the spectral content of input signals, this enhancement is selective for stimuli at low spatial frequencies and occurs irrespective of whether the luminance transients are actively generated or passively experienced. These findings indicate that, like eye movements, blinking acts as a computational component of a visual processing strategy that uses motor behavior to reformat spatial information into the temporal domain."

"The search for drivers of hominin speciation and extinction has tended to focus on the impact of climate change. Far less attention has been paid to the role of interspecific competition. However, research across vertebrates more broadly has shown that both processes are often correlated with species diversity, suggesting an important role for interspecific competition. Here we ask whether hominin speciation and extinction conform to the expected patterns of negative and positive diversity dependence, respectively. We estimate speciation and extinction rates from fossil occurrence data with preservation variability priors in a validated Bayesian framework and test whether these rates are correlated with species diversity. We supplement these analyses with calculations of speciation rate across a phylogeny, again testing whether these are correlated with diversity. Our results are consistent with clade-wide diversity limits that governed speciation in hominins overall but that were not quite reached by the Australopithecus and Paranthropus subclade before its extinction. Extinction was not correlated with species diversity within the Australopithecus and Paranthropus subclade or within hominins overall; this is concordant with climate playing a greater part in hominin extinction than speciation. By contrast, Homo is characterized by positively diversity-dependent speciation and negatively diversity-dependent extinction—both exceedingly rare patterns across all forms of life. The genus Homo expands the set of reported associations between diversity and macroevolution in vertebrates, underscoring that the relationship between diversity and macroevolution is complex. These results indicate an important, previously underappreciated and comparatively unusual role of biotic interactions in Homo macroevolution, and speciation in particular. The unusual and unexpected patterns of diversity dependence in Homo speciation and extinction may be a consequence of repeated Homo range expansions driven by interspecific competition and made possible by recurrent innovations in ecological strategies. Exploring how hominin macroevolution fits into the general vertebrate macroevolutionary landscape has the potential to offer new perspectives on longstanding questions in vertebrate evolution and shed new light on evolutionary processes within our own lineage."

T cells possess a vast diversity of surface receptors that bind to antigens presented on target cells, resulting in the activation of functions such as secretion of cytokines or cytotoxic molecules. T cell receptor (TCR) immunotherapies leverage this system to target tumor cells for elimination, yet methods of identifying rare TCRs remain nonspecific, resulting in many nonfunctional TCRs. We apply microcavity-containing hydrogel microparticles, known as nanovials, to selectively bind to and activate target T cells and capture secreted cytokines. This method enabled the linkage of TCR binding and functional secretion of cytokines directly with TCR sequences at the single-cell level, leading to expanded repertoires of TCRs and reduced false positives, ultimately enhancing the prospects of T cell cancer immunotherapy.
Abstract
The ability to selectively bind to antigenic peptides and secrete effector molecules can define rare and low-affinity populations of cells with therapeutic potential in emerging T cell receptor (TCR) immunotherapies. We leverage cavity-containing hydrogel microparticles, called nanovials, each coated with peptide-major histocompatibility complex (pMHC) monomers to isolate antigen-reactive T cells. T cells are captured and activated by pMHCs inducing the secretion of effector molecules including IFN-γ and granzyme B that are accumulated on nanovials, allowing sorting based on both binding and function. The TCRs of sorted cells on nanovials are sequenced, recovering paired αβ-chains using microfluidic emulsion-based single-cell sequencing. By labeling nanovials having different pMHCs with unique oligonucleotide-barcodes and secretions with oligo-barcoded detection antibodies, we could accurately link TCR sequences to specific targets and rank each TCR based on the corresponding cell’s secretion level. Using the technique, we identified an expanded repertoire of functional TCRs targeting viral antigens with high specificity and found rare TCRs with activity against cancer-specific splicing-enhanced epitopes."

Structured Abstract
INTRODUCTION
Drugs of abuse produce pleasurable feelings and reinforce consummatory behavior directed toward their acquisition. These same properties are characteristic of natural rewards that satisfy innate needs, such as food or water. Decades of research has shown that brain systems processing natural rewards are also impacted by drugs of abuse at the physiological, circuit, cellular, and molecular levels. These findings raise the hypothesis that drugs of abuse cause addiction by “hijacking” a common reward pathway, ultimately promoting drug intake while curbing other healthy goals. However, the specific neural substrates for such a shared reward pathway remain unidentified.
RATIONALE
Identification of a neural substrate that processes multiple classes of rewards necessitates multimodal analysis of neurobiological functions. This includes pinpointing central nodes that respond to reward exposure, examining specific cell types within this brain node that encode distinct rewarding experiences within the same individual, and identifying molecular effectors that mediate cellular and physiological adaptations. For this purpose, we employ a combination of approaches including whole-brain neuronal activity mapping, in vivo two-photon longitudinal calcium imaging at single-neuron resolution, and single-cell sequencing after in vivo CRISPR perturbation of a candidate gene. These multifaceted approaches enable the exploration of multiple components that comprise a common reward pathway and allow us to study how repeated drug exposure “hijacks” innate needs through this shared conduit.
RESULTS
Using whole-brain FOS mapping combined with chemogenetic inhibition approaches, we identify the nucleus accumbens (NAc) as a central hub necessary for both cocaine and morphine to disrupt natural reward (food and water) consumption. In vivo longitudinal tracking of individual dopaminoceptive neuron activity in the NAc in awake, behaving mice revealed overlapping ensemble responses across drugs of abuse and natural rewards, with drugs producing greater levels of activation. Repeated exposure to drugs of abuse augmented cell type–specific neural dynamics indicative of an escalation of drug responses, and subsequently disorganized natural reward processing in the NAc after drug withdrawal. We then developed a “FOS-Seq” approach to correlate brain-wide FOS patterns with brain-wide in situ gene expression data. We identified Rheb, a gene encoding a small GTPase that activates the mTOR pathway, as being correlated with FOS induction by chronic exposure to either cocaine or morphine. By integrating in vivo CRISPR perturbation of Rheb with single-nucleus RNA sequencing in the NAc, we demonstrated an essential role of Rheb in regulating signal transduction pathways associated with drug action in dopaminoceptive cells, and in diminishing natural reward consumption after chronic exposure to drugs of abuse. Finally, functional mapping of NAc-projecting neurons from regions that are activated by drugs of abuse points to orbitofrontal cortex as a potential ascending node that curbs natural reward consumption as verified with chemogenetic activation.
CONCLUSION
We delineated a common reward pathway that enables drugs of abuse to interfere with the fulfillment of homeostatic needs for food or water. These findings provide mechanistic insights into the intensification of drug-directed behavior in substance use disorders."
