Non chronic scratching a temporary itch may stimulate the immune system

Good news! But this research seems rather confusing or confused!

"... Researchers studied the effects of scratching by applying allergens to mice ears to cause a type of skin inflammation called dermatitis.
Some mice wore a tiny “cone of shame” to prevent scratching, while others were able to scratch their ears as they pleased.
The rodents that scratched showed more inflamed ears, with a higher immune response, but they also had less Staphylococcus aureus, a harmful bacteria known for causing skin infections. The study shows “that the act of scratching is really important” in producing more immune cells, wound healing ... With conditions that involve chronic itching, like psoriasis, scratching might only lead to further skin damage, the researchers say.

The study also dug into what goes on at the molecular level, finding previously unknown players in the itch-scratch cycle. As Aaron Ver Heul writes in a related Science Perspective, “beyond defining a previously unidentified neuroimmune itch-scratch circuit, the findings … may lay a foundation for discoveries to help people suffering from chronic itch.”"

INTRODUCTION
Scratching is an often irresistible, stereotypical, and evolutionarily conserved behavioral response to the sensation of cutaneous itch. In many common skin diseases, such as dermatitis (eczema), protracted itching—or pruritus—is the dominant symptom and represents a substantial source of morbidity. Scratching in response to itch is clinically well recognized to exacerbate dermatitis and is pathogenic in some diseases. However, scratching an itch is often a pleasurable sensation and does not trigger avoidance behavior, which suggests that it may provide some benefit to the host.
RATIONALE
In the skin, activated dermal mast cells mediate hallmarks of the cutaneous allergic response, including hives and itch, which results in scratching behavior. They also recruit inflammatory cells and promote host defense against Staphylococcus aureus. Mast cells can be activated through a variety of mechanisms, including allergens, that cross-link preformed complexes of immunoglobulin E (IgE) antibodies bound to the FcεRI receptor or via ligands for the MrgprB2 receptor, including substance P (SP)—a neuropeptide released by pain-sensing neurons. The functional consequence of mast cell activation through different receptors, how scratching promotes cutaneous inflammation, and whether scratching provides benefit to the host all remain poorly explored.
RESULTS
To explore the relationship between itch, scratching, and inflammation, we generated mice that allow for the selective and inducible ablation of the nonpeptidergic 2 (NP2) subset of itch-sensing neurons, characterized by the expression of MrgprA3 (called Mrgpra3DTR).
We found that MrgprA3-expressing neurons were required for scratching and inflammation in models of type 2 contact hypersensitivity and FcεRI-mediated mast cell activation. In both cases, scratching augmented mast cell degranulation, tumor necrosis factor (TNF) expression, and recruitment of neutrophils. Scratching was not required for increased expression of the alarmins thymic stromal lymphopoietin (TSLP) and interleukin-33 (IL-33), which are known to activate mast cells.
Rather, we found that scratching was sufficient to trigger release of SP from Trpv1-expressing neurons that synergized with FcεRI cross-linking, resulting in maximal TNF release from mast cells. This was confirmed using mice with a genetic ablation of MrgprB2 or the gene encoding SP (Tac1) and by chemogenetic inhibition of Trpv1-expressing neurons. Inflammation in mice prevented from scratching could be rescued by exogenous activation of Trpv1-expressing neurons. Finally, we found that scratching reduced cutaneous microbial diversity and, in an epicutaneous S. aureus infection model, both inflammation and host defense required scratching.
CONCLUSION
The itch-scratch cycle is a pathogenic process in allergic skin rashes, such as dermatitis, or arthropod reactions. In this cycle, itch and scratching increase inflammation and disease exacerbation.
Our data suggest that scratching activates cutaneous Trpv1-expressing neurons, which are a major source of SP in the skin. Coordinated activation of mast cells by both MrgprB2 and FcεRI agonism synergistically augments inflammation, in part through increased recruitment of neutrophils. Thus, dermal mast cells occupy a central node in cutaneous inflammation and are capable of integrating both adaptive and innate neuroimmune triggers.
Moreover, inflammation-induced scratching can reduce the abundance of certain members of the cutaneous commensal community and, in the context of superficial S. aureus infection, inflammation triggered by scratching provides enhanced host defense. These data exemplify how scratching can both exacerbate disease and benefit the host through a neuroimmune axis and reconciles the seemingly paradoxical role of scratching as a pathological process and evolutionary adaptation."

The epigenetic clock theory suggests that aging occurs due to the accumulation of epigenetic modifications, minor changes to the chemical structure of DNA that do not alter the underlying sequence, but instead change which genes are on or off. Unlike mutations, epigenetic modifications can also be reversed in some cases. ...

To answer this fundamental question, researchers analyzed data from 9,331 patients catalogued in the Cancer Genome Atlas and the Pan-Cancer Analysis of Whole Genomes. By comparing genetic mutations to epigenetic modifications, they found that mutations were predictably correlated with changes in DNA methylation, one type of epigenetic modification. They found that a single mutation could cause a cascade of epigenetic changes across the genome, not just where the mutation occurred. Using this relationship, the researchers were able to make similar predictions of age using either mutations or epigenetic changes. ...

In an analysis of multimodal data from 9,331 human individuals, we found that CpG mutations indeed coincide with changes in methylation, not only at the mutated site but with pervasive remodeling of the methylome out to ±10 kilobases.

Importance
Cannabis use has increased globally, but its effects on brain function are not fully known, highlighting the need to better determine recent and long-term brain activation outcomes of cannabis use.

Objective
To examine the association of lifetime history of heavy cannabis use and recent cannabis use with brain activation across a range of brain functions in a large sample of young adults in the US.

Design, Setting, and Participants
This cross-sectional study used data (2017 release) from the Human Connectome Project (collected between August 2012 and 2015). Young adults (aged 22-36 years) with magnetic resonance imaging (MRI), urine toxicology, and cannabis use data were included in the analysis. Data were analyzed from January 31 to July 30, 2024.

Exposures
History of heavy cannabis use was assessed using the Semi-Structured Assessment for the Genetics of Alcoholism, with variables for lifetime history and diagnosis of cannabis dependence. Individuals were grouped as heavy lifetime cannabis users if they had greater than 1000 uses, as moderate users if they had 10 to 999 uses, and as nonusers if they had fewer than 10 uses. Participants provided urine samples on the day of scanning to assess recent use. Diagnosis of cannabis dependence (per Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition criteria) was also included.

Main Outcomes and Measures
Brain activation was assessed during each of the 7 tasks administered during the functional MRI session (working memory, reward, emotion, language, motor, relational assessment, and theory of mind). Mean activation from regions associated with the primary contrast for each task was used. The primary analysis was a linear mixed-effects regression model (one model per task) examining the association of lifetime cannabis and recent cannabis use on the mean brain activation value.

Results
The sample comprised 1003 adults (mean [SD] age, 28.7 [3.7] years; 470 men [46.9%] and 533 women [53.1%]). A total of 63 participants were Asian (6.3%), 137 were Black (13.7%), and 762 were White (76.0%).
For lifetime history criteria, 88 participants (8.8%) were classified as heavy cannabis users, 179 (17.8%) as moderate users, and 736 (73.4%) as nonusers. Heavy lifetime use (Cohen d = −0.28 [95% CI, −0.50 to −0.06]; false discovery rate corrected P = .02) was associated with lower activation on the working memory task. Regions associated with a history of heavy use included the anterior insula, medial prefrontal cortex, and dorsolateral prefrontal cortex.
Recent cannabis use was associated with poorer performance and lower brain activation in the working memory and motor tasks, but the associations between recent use and brain activation did not survive false discovery rate correction. No other tasks were associated with lifetime history of heavy use, recent use, or dependence diagnosis.

Conclusions and Relevance
In this study of young adults, lifetime history of heavy cannabis use was associated with lower brain activation during a working memory task. These findings identify negative outcomes associated with heavy lifetime cannabis use and working memory in healthy young adults that may be long lasting."

"Evidence suggests that maternal health in pregnancy is associated with autism in the offspring. However, most diagnoses in pregnant women have not been examined, and the role of familial confounding remains unknown. Our cohort included all children born in Denmark between 1998 and 2015 (n = 1,131,899) and their parents. We fitted Cox proportional hazard regression models to estimate the likelihood of autism associated with each maternal prenatal ICD-10 diagnosis, accounting for disease chronicity and comorbidity, familial correlations and sociodemographic factors.
We examined the evidence for familial confounding using discordant sibling and paternal negative control designs.
Among the 1,131,899 individuals in our sample, 18,374 (1.6%) were diagnosed with autism by the end of follow-up. Across 236 maternal diagnoses we tested (prevalence ≥0.1%), 30 were significantly associated with autism after accounting for sociodemographic factors, disorder chronicity and comorbidity, and correction for multiple testing. This included obstetric, cardiometabolic and psychiatric disorders (for example, diabetes in pregnancy (hazard ratio (HR) 1.19, 95% confidence interval (CI) 1.08–1.31) and depression (HR 1.49, 95% CI 1.27–1.75)), previously shown to be associated with autism.
Family-based analyses provided strong evidence for familial confounding in most of the observed associations.
Our findings indicate pervasive associations between maternal health in pregnancy and offspring autism and underscore that these associations are largely attributable to familial confounding."

An all-optical method is demonstrated to detect and dynamically track the natural vibrational frequencies of a microorganism across the megahertz to terahertz spectral range. We uncover long-lived coherent oscillations in a single virus at room temperature that persist for many nanoseconds. These coherent signals give rise to an acoustic spectrum that is highly sensitive to the virus morphology and interactions between its glycoproteins and the environment. The methodology promises to shed light on viral dynamics without labeling and could serve as a means for viral fingerprinting.
Abstract
The natural vibrational frequencies of biological particles such as viruses and bacteria encode critical information about their mechanical and biological states as they interact with their local environment and undergo structural evolution. However, detecting and tracking these vibrations within a biological context at the single particle level has remained elusive.
In this study, we track the vibrational motions of single, unlabeled virus particles under ambient conditions using ultrafast spectroscopy. The ultrasonic spectrum of an 80 to 100 nm lentiviral pseudovirus reveals vibrational modes in the 19 to 21 GHz range sensitive to virus morphology and 2 to 10 GHz modes with nanosecond dephasing times reflecting viral envelope protein interactions. By tracking virus trajectories over minutes, we observe acoustic mode coupling mediated by the local environment. Single particle tracking allows the capture of viral disassembly through correlated mode softening and dephasing. The sensitivity, high resolution, and speed of this approach promise deeper insights into biological dynamics and early-stage diagnostics at the single microorganism level."

And now, despite all the subventions, it is shutting down about 15 years early as a monument to green fantasies financed with Other People’s Money, inflicted upon electricity ratepayers in California denied options to escape the madness engendered by “climate” hysteria supported by no actual evidence.

Ivanpah sits on 3471 acres of the Mojave Desert, with 347,000 mirrors controlled by computers, delivering concentrated sunlight into three towers. When fully operational, it was supposed to generate about 1 million megawatt-hours of power annually; for 2015–2023, actual annual output has averaged 702,322 MWh.

This poor performance has obtained despite a $1.6 billion loan guarantee from the Department of Energy, for which the plant owners did not have to pay even the usual credit subsidy cost (the expected default liability for the federal government), under the DoE section 1705 loan program. The plant owners then applied for and received a grant of $535 million from the Treasury Department under the section 1603 program to pay back part of the loan already guaranteed at no charge by the DoE. There also was the 30 percent investment tax credit, the accelerated depreciation (an assumed plant life of five years), and a depreciation bonus of 50 percent in the first year. And there is the guaranteed market share for wind and solar power—the “renewable portfolio standard”—which meant that the major California utilities were forced to buy the power produced by Ivanpah. (Pacific Gas & Electric buys the power from towers 1 and 3, while Southern California Edison buys the power from tower 2.) ..."

Despite its importance, the specific mechanisms driving this enhancement remain unclear.
In this study, we introduce a decomposition scheme based on the rank of irreducible tensors forming the 3NF, derived from chiral effective field theory at next-to-next-to-leading order, to elucidate their influence on SO splitting.
Within the shell-model framework, our analysis reveals that the rank-1 component of the 3NF is the primary factor enlarging the energy gap between the  and  single-particle levels in p-shell nuclei, while the rank-2 component makes a subdominant contribution.
Since the rank-1 component originates exclusively from the 2π-exchange 3NF, our finding will not depend on the choice of the low-energy constants of contact terms. We also remark on the antisymmetry of the rank-1 3NF, which can affect the quantum entanglement of spin states. This study lays the groundwork for further exploration into this field toward a microscopic understanding of the 3NF impact on the nuclear shell structure."
